At present, school, college, university going girls are often being hunted of eve teasing on the street.It is to say, hunting of eve teasing of a girl is so general visible, just in its opposition the visible of penalty of eve teaser is rare. Learner or not learner such woman as well take mentally preparation before going out from their room for this that she would be teased on the path with slung languages or bad attitudes by vagabonds. ‘Eve teasing’ on the street is one such reflection o f the inner beast in men, women are subjected to this social evil irrespective what background they are from. The lexically meaning of teasing is to make fun of a person playfully or annoyingly.It is a euphemism used in the sub continent for sexual harassment or modesty of women by men, with eve being a reference to the biblical Eve. Namely, staying hither and thither or turning point of college going path, to make tease known or unknown girl, or make tone, comment or display abetment or throw slang language or attempt to capture photography by mobile phone etc is called eve teasing, which includes a small sexual harassment or rape. If there is no parent or guardian with any woman who will be hunted of eve teasing, we can say that definitely. Now think a bit, we can give hundred percent surety concerning eve teasing.But can we give one percent as to any positive deed? All suicides caused by eve teasing do not come to light on the news paper. In addition to last some years, Trisha, R umi, shimi, Sadia, Toshi such many unknown mothers-sisters have departed from this beau world. Question is here-why did they give their lives? Did they have no right to protest, move freely and live fairly? Definitely, they had said rights as per Article-11, 32 and 36 of Bangladesh Constitution which denote as fundamental rights to live in the society free and fairly. But Allah says in the Holy Quran- Man is the protector of the Woman. The question is that, why do they keep silent. The answer is that, they have no might and no body to stand beside them. Nevertheless some so-called reformists talk concerning women’s rights. Here there is apprehension that they who talk regarding woman’s rights, infringe at first from their family. In the name o f woman’s rights or modern era they would like to bring out the on road and set up the western culture in this country.Someone claim that Islam dishonors woman and treat her as inferior and they have no equal position in society like man. Bacterial Transformation Using pGLO Involving X and Y Genes

Genetic transformation is due to a direct cause in the change by genes, due to the cell in taking and expressing traits from a separate piece of DNA. Naturally proficient bacteria are able to absorb exogenous DNA and go through genetic transformation. (Chen & Dubnau, 2004) The purpose of this experiment was to discover how a gene could be moved from one organism to a different organism with the help of plasmid. The cells that are capable of acquiring these traits from the other organism are known as being competent.Weedman, 2013). In this particular experiment we will genetically transform the bacteria E. coli by inserting a gene through heat shock, this gene codes for Green Fluorescent Protein, also known at GFP. The GFP gene originally comes from a Jellyfish and under an ultraviolet light the bacteria that acquired the gene with glow a brilliant fluorescent green color. (Portman et al. 2013). If the cells' nutrient medium has the sugar arabinose added to it then GFP can be turned o n. (Weedman, 2013). To determine if our hypothesis was correct, we used four differently prepared plates.The four plates each contained a different combination of the following; arabinose, ampicillin, LB nutrient broth, and pGLO plasmid. The combinations were; +pGLO LB/amp, +pGLO LB/amp/ara, -pGLO LB/amp, and -pGLO LB. Our hypothesis was: the plates with pGLO will have growth because they are resistant to the antibiotics involved, the plate with ampicillin and without pGLO will show no growth due to the fact that the antibiotic compromises the bacteria, and the plates that will grow will be the ones containing pGLO since they obtain the trait for glowing.Materials and Methods: All methods were obtained from (Weedman, 2013) Before beginning the experiment obtain latex gloves, two microcentrifuge tubes, a beaker filled with ice, a micropipetter, micropipetter tips, transformation solution containing calcium chloride, sterile loops, pGLO, E. coli, and four plates containing different s ubstances. To begin label the two microcentrifuge tubes +pGLO and – pGLO. Then proceed to obtain 250ul of transformation solution and put it in each one of the tubes using a different miropipetter tip each time, this solution will help enhance the permeability of the cell membranes.Then use a sterile loop to acquire single colony of E. coli to add to the tube labeled +pGLO; add this by twisting the sterile loop until the pGLO is off. Then repeat the last step for the -pGLO tube using a new sterile loop. Next add pGLO to the tube labeled +pGLO, to do this take a new sterile loop and inserted it into a vile containing the plasmid pGLO. Then twist the loop into the tube labeled +pGLO, then place both tubes into the beaker filled with ice for approximately 10 minutes. While the tubes are on ice grab the four LB (Luria Bertani broth) nutrient agar plates.Each plate should be labeled either +pGLO or – GLO; you should nave 1 LB/amp/ara plate (+pGLO), 1 LB plate (-pGLO 2 LB/am p plates (+pGLO)(-pGLO). After 10 minutes in the ice bath place the tubes in a floating rack and put them in a 420C water bath for exactly 50 seconds, giving them a heat shock. Immediately place both tubes back in the ice after the water bath for approximately 2 minutes. Once 2 minutes is up remove the tubes from the ice and put them in the rack at room temperature. Using a new tip each time, add 250ul of nutrient broth to both tubes. Then close the tubes and let them sit at room temperature for 10 minutes.After 10 minutes flick both tubes with your fingers to ix the contents, then using a fresh tip each time add 100ul of the transformation solution (+pGLO) and the control (-pGLO) to their appropriately labeled plates. Using a new sterile loop each time spread the contents around in each dish. Then tape the plates together and placed them upside-down in an incubator set at 370 C for 24 hours. Results: This experiment shows how a gene can be transferred from one organism to a differe nt organism through the help of plasmid. Traits are exchanged from one DNA stand toa different one in the bacteria E. coli.Two of the plates were a control group, hich meant there was no growth after the plates were taken out of the incubator. These two control plates were the ones containing -pGLO LB/amp and -pGLO LB. The transformation plates were the two plates containing +pGLO LB/amp and +pGLO LB/ amp/ara. These two plates showed a substantial growth in bacteria after being taken out of the incubator, one plate showing a considerably larger growth than the other and they both glowed under UV light due to the pGLO. Michael Gregory did a previous experiment; he came to the same conclusion that our experiments' results oncluded.His experiment was identical to ours, involving the same materials and procedure. The same plates showed growth in his experiment as ours, as well as the plates that didn't show growth were the same. (Gregory, 2004). The only weakness that I could think of that would have a major effect on the results would be not using sterile equipment and causing cross contamination. Our experiments did not have any problems arise that would affect the results we obtained.

He outwardly struggles to find his place and role in the society even as he struggles inwardly to understand his own nature. As Yoda and Obi-Wan explained to him, Luke must confront and go beyond the dark side to be a Jedi. He is confronted with facing Darth Vader, who he has just learned to be Anakin Skywalker—his father, and in whom Luke believes still has good in him and who he says he could not kill. He was also confronted with troubles that has shaped him into the peson he chose to be. Vader has been a Jedi once, turned to the dark side of the Force because he could not accept the death of his loved ones—first of his mother, then of his intuition that had foretold the death of his wife, Amidala. During their fight, he tried to convince his son that turning to the dark side was the only way Luke could save his friends. Luke was like his father, reckless in his actions and impatient in his youth. But as he matured, he learned the true path of the Force, with the guidance of Yoda and Obi-Wan. Like his father before him, he, too, was troubled with death and suffering. Yoda, sensing death coming to him, explains to Luke: â€Å"Strong am I with the Force, but not that strong. Twilight is upon me and soon night must fall. That is the way of things, the way of the Force.† Inevitably at Endor, Luke has come to confront his father, not with the intention of fighting him as before, but to convince him to turn back to the good side. His attempts turns out to be futile. He further exhibits his maturity from being reckless and impatient by restraining himself when given the chance to strike at the Emperor. However, the Emperor seduces Lukes hatred and anger to attack him. Luke is tormented but still restrains himself until finally his rage erupted and attacked the Emperor. Darth Vader responded in defense of his master and Luke ends up fighting with his father. Luke realized that the Emperor was using his rage to turn him to the dark side of the Force. He has defeated Vader, not out of some personal, revenge-driven desire, but in order to protect someone he loves—Vader has treathened Lukes newly found twin sister Leia. As he looked down on his father with pity and understanding, he realized that he, too, has the potential to turn to the dark side. He threw away his lightsaber in his firm belief that he could still save his fathers soul and save himself from the path of the dark side by not killing Vader. By doing so, he has rejected evil in himself and submits himself to the will of the Force. As Yoda explained to him: â€Å"A jedis strenght flows from the Force. But beware. Anger, fear, agression, the dark side are they. Once you start down the dark path forever will it dominate your destiny.† He has faced his own fears and defeated them, evidenced by his reply to the Emperors offer to take his fathers place at his side: â€Å"Never. Ill never turn to the dark side I am a Jedi, like my father before me.† He has set an example that evil does not come naturally from the troubles that we are faced with, and that being good or evil is a matter of choice.

Another limitation is the difficulty in estimating the age of things which are older than 50000 years as the amount of C-14 in such samples become absolutely undetectable due to complete decay. Recently, bravery and courage on the battlefield, has given him the privilege to return home; he’s been told to report at headquarters at 0700 hours the next day. Transported in a truck by a fellow officer, he learns he is officially relieved of his duties in Iraq after his one last mission to bring, from Baghdad, a convoy of four hummers, two trucks, and two support helicopters into Kuwait. Days later he soon found himself traveling by train, boat, and plane to finally reach his final destination, New York City. Vincent was greeted with a halo of happiness as his family came to bring him home in their Ford F-150. Imagine now, all these vehicles, the cars, trucks, helicopters, trains, boats, planes are no longer fueled by petroleum oil, but from bio-diesel oil. Amazingly, bio-diesel is no longer an imagination, but a close reality as this renewable fuel is vastly becoming the next best alternative source for replacing the old and diminishing fossil fuels. The soybean plant has been apart of US history for decades as an important product of American agriculture. Today, soybeans are Americas’ number one export crop, and for nearly 30 years beginning in the 50’s, the US was producing about 75 percent of the worlds soybean crop (Soypower). The oil that is extracted from this simple bean has found its way into some of our common household products, such as shortening and salad dressings. Weight-loss products, baby formulas, and sports drinks also contain soy proteins, which provide us all nine essential amino acids. Even with all the important and interesting capabilities the soybean has been able to bring to us, none may be as significant as biodiesel fuel that some have been raving about. â€Å"Is it compatible with current technology?† I believe an important and very relevant question to be answered before ever implementing an invention like bio-diesel oil into society is: A computer becomes more dynamic every year, but it is still designed in such a way to be compatible with floppy disks, zip disks, CD’s, and now, the Flash chips. The same must be true for bio-diesel oil or a very complicated issue would be at stake. Thankfully, for us, bio-diesel oil has been tweaked in such a way that petro-diesel oil and bio-diesel oil could be used simultaneously in practically every current diesel engine there is.

So far six actors have taken on the role of James Bond and all were significant super fit good-looking and popular actors. This only helps to increase the popularity of the films and now that the cinema is public it means people of all ages go to watch these films with family and friends. Many films and actors have taken on similar storylines and characters to that of the Bond films such as Johnny English and the Austin Powers: International Man of Mystery films put a lighter, humorous side to the real life dangers that spies face in reality. Also films such as the Alex Rider books based Stormbreaker and the agent Cody Banks films attract a different audie nce than the Bond films in a way because they use young actors and actresses to put across a more understandable and sometimes more watchable film for the younger audience. The Mission Impossible films were based on a 1960’s television show of the same name. Tom Cruise would have been deliberately chosen to take the lead character role in these films. Like the Bond actors this increases interests in the film and also makes the general view and activities of espionage seem glamorous but yet action packed. Many of the signs such as the gunshots, explosions and police sirens make these films suspense action thrillers. I think that the way that the directors use the certain signs to help glamorise the life of spies is clever and very appealing to the audience. The Harlem Drag Culture Foucault, Moraga, Fuss, hooks, Butler. These authors, along with many more, have concerned themselves with the defining of categories. In reading these authors our class has, upon every occasion of meeting, discussed the formation of categories. What we have discovered, in part, is that things are not defined by what they are, but by what they are not. Diana Fuss, in her article "Inside/Out," states "any identity is founded relationally, constituted in reference to an exterior or outside that defines the subject's own interior boundaries and corporeal surfaces" (Fuss, "Inside/Out," 234). A common example of this, derived from Freud, is that males are defined by their having a penis, while females are defined by their lack of one. Defining identity is not necessarily so binary. As Cherrà ­e Moraga puts it "Call me something meant to set me apart from you and I will know who I am" (Moraga, "The Breakdown of the Bicultural Mind," 237). The subcultures of American society, cultures that are not part of the white patriarchy, are defined by how they differ from this white patriarchy. Our class was privileged to be introduced to one subculture found in Harlem during the late 1980s through the documentary Paris is Burning by Jennie Livingston (1992). This documentary captured the lives of men who lived outside the dominant culture. They had several strikes against them: they were Latino and African American, they were homosexual, and many of them were poor, sometimes even homeless. These men came together to form a kinship network, in the form of houses, to protect and support one another. Out of this milieu developed drag balls, balls in which the men dressed up and competed in different categories, such as "executive," "realness," and "voguing." The object of most of these categories was to mimic dominant society by looking like the heterogeneous members of the white patriarchy. After watching Paris is Burning, reading critiques of it, listening to class discussions, and processing through my own thoughts about the film that I have come to struggle with an extremely large tension I am confronted with in thinking about the drag ball subculture of Harlem. Is the mimicking of dominant society by this culture a way of subverting it or is it supporting and perpetuating the white patriarchal ideal? Do these men redefine dominant society in their own terms and take control of it?

The Fisherman by Kurt Brown

Life is filled with pleasures and uncertainties. We have ups and downs, and enjoy the better days because we have survived the worst. The â€Å"Fisherman† by Kurt Brown, tells a tale of a man fishing every day in hopes of catching something beyond him. This poem represents our lives and is a direct metaphor of the duality of success and failure. Our lives can relate to this poem because there comes a time in our lives, when we realize the world has so much to offer us. In order to see and feel the many wonders in life, risk needs to be put forth.Life does do not come to us, like the fisherman; who continues to cast his line into the deep, dark, and thundering waters, perseverance is required. Someday the catch will be rewarding and others will leave empty handed. However, it was enjoyable to go out and risk catching something glorious. Fishing is sport that involves any body of water. Theses body of waters are always bigger than our self’s. â€Å"Maybe it's the shadow of other fish, greater than his, the shadow of other men's souls passing over him,† (Brown 6,7) after analyzing this line, I concluded Brown was referencing life and how it continues without us.There is always a Goliath in life that towers over us all. However, we should not let it pass without attempting to grasp the monstrosity that is life. Not all catches are easy; sharks, marlin and other big game fish will all struggle to pull you down with them into the depths of the deep, dark Ocean. Once you hook the fish, a fight is ensured that will test the limits of your heart. Whether it is with bigger fish or bigger men, strength is required to overcome them. I believe this is one message brown intended to convey in his poem.In a way, Brown tells an aspiring tale of the fisherman's success and failures. â€Å"Each day he grabs his gear and makes his way to the ocean,† (Brown 8,9) determined to catch the best in the salty waters, yet satisfied knowing if he goes home empty handed, the best of him was put forth. Brown was trying to showcase the relentlessness of the fisherman's pride. Each day was different and offered a better catch than before, even if nothing as caught in the previous days, the fisherman continued to have faith and cast his line.In life, we feel like this. Some days we feel like all the choices made up to this point have been pointless. Then we remember, after time spent sulking about our mistakes, that we still have a chance our luck and livelihood. In a way, we all have a little fisherman in all of us. Failure in life is expected, like death, everyone has to face it head on. However, others charge right into it, hoping for the best outcome. We like to think control is always in our hands and the outcomes in majority of situations will tip towards our favor.However, all we have to rely on is uncertainty. Yes we can plan the lives we wish to live out, pursue the goals in life that will allow happiness to flow, but the only real goa l we all achieve knows that uncertainty reigns supreme. â€Å"He'll go back soon, thinking: â€Å"Maybe tonight. † (Brown 17) Brown conveyed the doubt that plagues the fisherman's conscious. â€Å"Maybe tonight,† he is unsure whether the trip back is worth it. Although he may be optimistic and determined, he can't shake the uncertainty.Like a double-edged sword, you cannot have one without the other. In this case, it's doubt that attacks his unyielding perseverance. In closing, despite all of the contrasts of success and failure, Brown started his poem with a strong message â€Å"A man spends his whole life fishing in himself for something grand,† we all want to be the best versions of ourselves. But the thing is; if allowed, we get better as time goes on. We become wiser, and look back on all of the great things that we allowed ourselves to experience.So, to continue becoming grand, we have to constantly pull the greatness out of our souls and temper it. Like a blacksmith creating a fine piece of metal from raw material, it takes time, work, and fire to create a truly fine piece of work. We must constantly fuel the fire and if that fire should go out, it must be relight within to continue on. Like the fisherman in Brown's poem, preparation is required to fish properly. Otherwise, if a strong fish comes your way, how do you expect to make it yours?

Instead, these non-participants may abandon the purchase completely or fulfill the transaction in an offline setting.
